Tunnel Hill to Robards

KY 416 begins at an intersection with U.S. Route 41 Alternate (US 41 Alt.) in Tunnel Hill, within the south-central part of Henderson County. It travels to the east-southeast and intersects the southern terminus of KY 1217 (Pruitt Agnew Road). Immediately afterward, it crosses over the West Fork Canoe Creek and then a branch of that creek. It curves to the northeast and intersects the southern terminus of KY 1299. It intersects KY 283 on the western edge of Robards. After the highway enters Robards proper, it crosses over some railroad tracks of CSX. It then curves to the northeast and intersects US 41 on the northeastern edge of the city.

Robards to eastern terminus

KY 416 travels to the east-northeast and has an interchange with Pennyrile Parkway (future Interstate 69 (I-69)). Almost immediately, it intersects the northern terminus of KY 2096 (Alcan–Aluminum Road). It winds its way to the northeast and enters Niagara, where it has a one-block concurrency with KY 136. KY 416 crosses over Cash Creek and enters Coraville, where it intersects the eastern terminus of KY 520. It crosses over Lick Creek. It curves to the east-northeast and has a short concurrency with KY 1078. The highway then has an interchange with the Audubon Parkway (future I-369). The highway heads to the north-northeast and curves to the northwest. It curves to the northeast and passes a Patriot Coal mine. The highway then enters Hebbardsville, where it intersects the eastern terminus of KY 351 (Zion Road). At this intersection, KY 416 turns right and travels to the southeast. It continues to the southeast and meets its eastern terminus, a dead end on the western shore of the Green River.
