DeMelo played four seasons (2009–13) of major junior hockey in the Ontario Hockey League (OHL), scoring 25 goals and 100 assists for 125 points, while earning 220 penalty minutes in 218 OHL games played.

On April 20, 2012, DeMelo was signed by the San Jose Sharks of the NHL to an entry-level contract. He made his NHL debut for the Sharks on October 17, 2015 in a game against the New York Islanders. He scored his first NHL goal as a San Jose Shark on January 26, 2016 in a 6–1 win over the Colorado Avalanche.
