Lesla-Lar is a scientist in the bottle city of Kandor, and is the counterpart of Kara Zor-El from the Earth-Three universe, brought there by Jur-ll who was the father of Lesla's cousin Ultraman. Jealous of Supergirl's fame and life, Lesla-Lar invents a device that causes Supergirl to lose her powers. Now a normal human, Supergirl takes on her secret identity of Linda Lee full-time and is adopted by Fred and Edna Danvers. Lesla-Lar then uses another device to switch places with Linda, and assume the role of Supergirl for herself. She is able to convince Superman that she is Supergirl, and secretly meets with Lex Luthor to convince him to kill the Man of Steel. Luckily, Krypto the Superdog realizes that Lesla-Lar is an imposter, and manages to send her back to Kandor. In Action Comics #282 (November 1961), Linda's powers are restored by Mister Mxyzptlk; Lesla-Lar is arrested by the Kandorian police and her power sapping machine is destroyed.

Lesla-Lar returns in Action Comics #297 (February 1963). This time she switches places with Lena Thorul, the sister of Lex Luthor, who is also a double for Supergirl. She releases three Kryptonian criminals from the Phantom Zone and enlists their help to recover a cache of forbidden Kryptonian weapons. After finding the cache, one of the criminals, Kru-el, tests one of the weapons on Lesla-Lar, and disintegrates her.

Lesla-Lar's final appearance is in Superman Family #206 (March–April 1981). After her body was disintegrated by Kru-el, her intelligence managed to survive in a disembodied state. The process has affected her sanity, as she now believes she and Supergirl are sisters. After several unsuccessful attempts to destroy Supergirl by mentally influencing other villains against her and then getting trapped in a Superboy robot, Lesla-Lar invades Supergirl's mind and temporarily manages to expel Supergirl from her body. Supergirl's foster parents, Fred and Edna Danvers, confront Lesla-Lar and tell her they are convinced she is not their real daughter. The blow to Lesla-Lar's confidence is enough to allow Supergirl to reenter her body and force out Lesla-Lar. Lesla-Lar's mental energies are dispersed on the astral plane.
