The 2016–17 East Superleague is the fifteenth Superleague competition since the formation of the Scottish Junior Football Association, East Region in 2002. The winners of this competition are eligible to enter the 2017–18 Scottish Cup. The season began on 6 August 2016.

Member clubs for the 2016–17 season

Bonnyrigg Rose Athletic are the reigning champions.

St Andrews United and Tayport were relegated to the East Premier League at the end of the 2015–16 season and are replaced by Jeanfield Swifts and Dundonald Bluebell.

Lochee United were also promoted from the East Premier League after defeating Sauchie Juniors 10–0 on aggregate in the East Region Super/Premier League Play Off.
