Pgrac is an artificial promoter consisting of Bacillus subtilis groE promoter, lac operator and gsiB ribosome binding site. This is a strong IPTG-inducible promoter for B. subtilis, which was first appeared on plasmid pNDH33[1] and later on pHT01 of the company MoBiTec [2]. Plasmid pHT01 is supposed to be stable in both B. subtilis and Escherichia coli, and can be used for expression of proteins in these host strains. Using pNDH33/pHT01 proteins could be produced up to 16% of total protein in B. subtilis. Pgrac100 is an improved version of Pgrac, which can produce up to 30% of total cellular proteins in B. subtilis.
