The Duomo di San Primiano is a Roman Catholic cathedral located in Lesina, region of Apulia, Italy.

A church at this site was likely present from the medieval period. Rebuilt over the centuries, the church was destroyed by an earthquake in 1630. By the 1650s, another had been rebuilt, and this one replaced in 1828-1837. The roof fell in 1922, and was rebuilt in the 1950s. The church has a single nave with two lateral chapels. The church has frescoes depicting the Life of Christ by Bocchetti Gaetano.
