Roger Federer was the two-time defending champion, but lost in the semifinals to Novak Djokovic.

Djokovic won in the final 4–6, 6–4, 6–3, 7–6(7–2), against unseeded Jo-Wilfried Tsonga. It was Djokovic's first Grand Slam title of his career, and first of six Australian Open titles. It was also the first Grand Slam final since the 2005 Australian Open that did not feature either Federer or Rafael Nadal, who was beaten by Tsonga in the semi-finals.
