Maria Sharapova was the defending champion, but she was defeated in the third round to thirtieth seed Agnieszka Radwańska.
Justine Henin claimed the title without dropping a set, and defeating 2004 champion Svetlana Kuznetsova in the final, 6–1, 6–3. This would be her last Grand Slam title, and as well as her final US Open appearance.
Future champion and World No. 1 Angelique Kerber appeared in the main draw of the US Open for the first time. She lost to Serena Williams in the first round.
