Reigning champion Maria Bueno (BRA) did not defend her title, as she was suffering from jaundice. Seventh-seeded Angela Mortimer (GBR) defeated sixth-seeded Christine Truman (GBR) 4–6, 6–4, 7–5 in the final to win the Ladies' Singles tennis title at the 1961 Wimbledon Championships. It was the last all-British final to date.
